Pharmacological activity arises from agonism at the glucagon-like peptide-1 receptor, a G protein-coupled receptor expressed in the pancreas, the gastrointestinal tract, and the brainstem. Receptor activation raises intracellular cyclic adenosine monophosphate and enhances insulin release in a glucose-dependent manner, an effect that diminishes when blood glucose concentration is low. Other effects include slowed gastric emptying and hypothalamic satiety signalling. These pathways are described well. Receptor desensitisation rates across tissues, relative to the endogenous hormone, are still under investigation, and reported findings differ between laboratories.
The company that developed the compound filed it as a long-acting analogue, and it gained first approval in 2017 for type 2 diabetes. Later authorisations from several regulators extended the indication to chronic weight management, and the World Health Organization added the glucagon-like peptide-1 receptor agonist drug class to its model list of essential medicines in 2023. Production uses solid-phase peptide synthesis followed by side-chain conjugation and chromatographic purification. Supply constraints and cost differences across regions are well documented. Literature on long-term outcomes continues to grow, with many trials reporting surrogate endpoints rather than hard clinical endpoints.
Semaglutide is a synthetic peptide of thirty-one amino acids that shares roughly ninety-four percent sequence identity with human glucagon-like peptide-1. Two substitutions resist enzymatic cleavage by dipeptidyl peptidase-4, and a fatty diacid side chain attached through a linker promotes binding to serum albumin. That albumin binding slows renal clearance and extends the circulating half-life from minutes to approximately one week. The structural changes are well established in the published literature. Whether the same modifications affect receptor signalling bias in ways that matter clinically remains an open question.
Lyophilized peptide material is typically stored at or below -20 °C, with -80 °C used for longer-term archives. Vials should remain sealed and desiccated because moisture promotes aggregation and hydrolysis. Repeated freeze-thaw cycles are avoided since they can alter peptide conformation and reduce recovery. Once reconstituted, solutions are generally kept at 2-8 °C and used within a defined window. Stability beyond those windows depends on buffer composition and concentration, and exact limits are product-specific rather than universal.
Identity and purity are assessed with reversed-phase high-performance liquid chromatography, which separates the peptide from related impurities by hydrophobicity. Mass spectrometry confirms molecular weight and detects truncation or modification products. Peptide mapping after enzymatic digestion verifies the amino acid sequence. Quantitation is often performed by LC-MS/MS or by immunoassay, and the two approaches can give different values because they measure different things. Method validation parameters such as accuracy, precision, and limit of quantitation are reported alongside results.
Certificate of analysis documents from suppliers typically report purity by chromatographic area, water content, and counter-ion identity. Independent verification is advisable because reported values can be generated under differing conditions. Impurity profiles matter for research use, where aggregates, deamidation products, and residual solvents may influence experimental results. Container, lot, and chain-of-custody records support traceability. Analytical results are method-dependent, so comparisons between laboratories require the same procedure and reference standards.

Serum protein binding dominates the pharmacokinetic profile. The attached chain associates strongly with albumin, shielding the peptide from enzymatic attack and slowing filtration by the kidney. This interaction extends the circulation half-life to roughly one week in humans, which supports weekly administration intervals. An oral version pairs the peptide with an absorption enhancer that transiently alters gastric epithelium, permitting limited uptake; bioavailability by that route is substantially lower than by injection.
Semaglutide belongs to the glucagon-like peptide-1 receptor agonist class, a group of synthetic peptides that imitate an incretin hormone released by intestinal L cells after food intake. Native GLP-1 circulates for only a few minutes because dipeptidyl peptidase-4 cleaves it rapidly. The hormone acts on pancreatic islets, the gastrointestinal tract, and several brain regions. Because the natural peptide is short-lived, development work concentrated on analogues that keep receptor activity while resisting enzymatic breakdown and renal clearance.
The semaglutide sequence is a 31-residue analogue of human GLP-1, altered at three positions relative to the parent hormone. Aminoisobutyric acid replaces alanine at position 8, arginine replaces lysine at position 34, and a lipophilic diacid is attached to lysine 26 through a short linker. These features are reported consistently in the structural literature. The position 8 substitution blocks recognition by dipeptidyl peptidase-4, while the attached chain drives strong, reversible association with a carrier protein in blood.
Semaglutide dissolves readily in water and in aqueous buffers near neutral pH. Solubility decreases near the isoelectric point, where net charge is minimal. Common laboratory solvents include phosphate-buffered saline and dilute ammonium bicarbonate. Strongly acidic or basic conditions may accelerate hydrolysis. Working concentrations are usually prepared by diluting a concentrated stock. Vial surfaces can adsorb small amounts of peptide at low concentrations, so carrier proteins or low-binding tubes are sometimes used.
Reverse-phase high-performance liquid chromatography is the standard method for purity assessment, separating the peptide from truncated or oxidized variants. Mass spectrometry confirms molecular mass and detects modifications, while ultraviolet absorbance near 280 nanometers supports concentration measurement through tryptophan and tyrosine residues. Circular dichroism can indicate secondary structure, though the peptide is largely helical in solution, and ion-exchange chromatography resolves charge variants. Purity values above 95 percent are typical for research-grade material. Stability studies track degradation over time under defined conditions.
Lyophilized semaglutide is typically stored at temperatures between minus 20 and minus 80 degrees Celsius for long-term preservation. Short-term storage at 2 to 8 degrees Celsius is common for working aliquots. Repeated freeze-thaw cycles can degrade the peptide and are usually avoided. The molecule is hygroscopic in its solid form, so containers should remain sealed with desiccant. Solutions are less stable than powders and are generally prepared fresh. Light exposure is limited because aromatic residues can undergo photo-oxidation.
It mandates that "the use of artificial intelligence efforts should not compromise the integrity of nuclear safeguards, whether through the functionality of weapons systems, the validation of communications from command authorities, or the principle of requiring positive human actions in execution of decisions by the President with respect to the employment of nuclear weapons." In February 2026, the Trump Administration publicly reaffirmed that nuclear weapons decisions will remain subject to human control, with a senior Pentagon official reiterating the "Department's policy that there is a human in the loop on all decisions on whether to employ nuclear weapons". In September 2025, the French Center for AI Safety (CeSIA), The Future Society and the Center for Human-Compatible AI (CHAI) published a global call for AI red lines urging governments to reach a binding international agreement prohibiting unacceptable AI uses by the end of 2026. The declaration was initially signed by 200 prominent figures including 10 Nobel Prize winners, and was announced by Maria Ressa at the United Nations General Assembly. In December 2025, President Donald Trump signed an executive order to establish a "National Policy Framework for Artificial Intelligence". The executive order discouraged state governments from regulating AI, urging Congress to pass a law pre-empting such regulations. The White House cited economic and national security concerns as reasons for the measure, while some criticized Trump for creating uncertainty for AI regulation. On July 6, 2026, Illinois Governor J.B.
An induction dose of thiopental wears off after a few minutes because the thiopental redistributes from the brain to the rest of the body very quickly. Also, thiopental has a long half-life, and it needs time for the drug to be eliminated from the body. If a very large initial dose is given, redistribution is minimal because the body is saturated with the drug; thus, recovery of consciousness requires the drug to be eliminated from the body. Because this process is not only slow (taking many hours or days) but also unpredictable in duration, barbiturates are unsatisfactory for the maintenance of anesthesia. Thiopental has a half-life around 11.5 hours (but the action of a single dose is terminated within a few minutes by redistribution of the drug from the brain to peripheral tissues), and the long-acting barbiturate phenobarbital has a half-life around 4–5 days. In contrast, the inhaled anesthetics have extremely short half-lives and allow the patient to wake up rapidly and predictably after surgery. The average time to death once a lethal-injection protocol has been started is about 7–11 minutes. The thiopental takes only about 30 seconds to induce anesthesia, the pancuronium takes 30–45 seconds to cause paralysis, and the potassium takes about 30 seconds to stop the heart, so death can theoretically be attained in as little as 90 seconds. Given that it takes time to administer the drug, time for the line to flush itself, time for the change of the drug being administered, and time to ensure that death has occurred, the whole procedure takes about 7–11 minutes.
Jeffrey Robert Bagwell (born May 27, 1968) is an American former professional baseball player and coach. A first baseman, he spent his entire 15-year Major League Baseball (MLB) playing career with the Houston Astros. Originally a Boston Red Sox fourth-round selection from the University of Hartford in the 1989 amateur draft, Bagwell was traded to the Astros in 1990. Bagwell was named the National League (NL) Rookie of the Year in 1991 and won the NL Most Valuable Player Award (MVP) in 1994. Bagwell and longtime Astros second baseman Craig Biggio and teammate Lance Berkman were known as the "Killer B's", and the team experienced consistent success during their careers; Houston finished in first or second place in the NL Central division in eleven of twelve seasons from 1994 to 2005. During that period, the Astros qualified for the playoffs six times, culminating in Bagwell's lone World Series appearance in 2005. Bagwell hit 449 home runs for the Astros, the most in club history, and set numerous other franchise career and single-season records. He is a four-time MLB All-Star, a three-time Silver Slugger winner and a Gold Glove recipient. The only player in MLB history to have six consecutive seasons (1996–2001) with thirty home runs, 100 RBIs, 100 runs scored, and 100 walks, Bagwell is one of twelve players in history to hit 400 home runs and record an on-base percentage (OBP) of .400. He is the only first baseman with at least 400 home runs and 200 stolen bases.

== Prevalence == Comorbidity of addictive disorders and other psychiatric disorders, i.e., dual disorders, is very common and a large body of literature has accumulated demonstrating that mental disorders are strongly associated with substance use disorders. Adolescents and young adults are particularly at risk for dual diagnosis, as early substance use can interfere with brain development and exacerbate emerging mental health conditions. The 2011 USA National Survey on Drug Use and Health found that 17.5% of adults with a mental illness had a co-occurring substance use disorder; this works out to 7.98 million people. Estimates of co-occurring disorders in Canada are even higher, with an estimated 40-60% of adults with a severe and persistent mental illness experiencing a substance use disorder in their lifetime. A study by Kessler et al. in the United States attempting to assess the prevalence of dual diagnosis found that 47% of clients with schizophrenia had a substance misuse disorder at some time in their life, and the chances of developing a substance misuse disorder was significantly higher among patients with a psychotic illness than in those without a psychotic illness. Another study looked at the extent of substance misuse in a group of 187 chronically mentally ill patients living in the community. According to the clinician's ratings, around a third of the sample used alcohol, street drugs, or both during the six months before evaluation. Further UK studies have shown slightly more moderate rates of substance misuse among mentally ill individuals.
=== Organic reactions === Hydrogen peroxide is frequently used as an oxidizing agent. Illustrative is oxidation of thioethers to form sulfoxides, such as conversion of thioanisole to methyl phenyl sulfoxide:
== Research == Moroder started his peptide research with the synthesis of the S-peptide of ribonuclease A and studies on this protein-peptide complex. It was one of the first demonstrations of the key and lock principle in peptide hormone receptor interactions. As research associate he worked on the synthesis of radioactive adrenocorticotropin, which represents one of the first synthetic research works on human peptide hormones. Moroder's work at the Max Planck Institute for Biochemistry in Martinsried was initially focused on the gastrin and cholecystokinin system, revealing the mechanism for the membrane-bound pathway of hormone recognition by the receptors. In parallel, he worked on synthetic methods in peptide and protein chemistry such as the introduction of di-tert-butyl dicarbonate as a general and widely used reagent in peptide chemistry, regioselective assembly of cystine-rich peptides, and the synthesis of highly robust disulfide and diselenide scaffolds. In the later phase of his research, Moroder became increasingly interested in the study of more complex biological and medical systems by chemical means. For example, he addressed fundamental questions of the kinetics of protein folding and actively contributed to the design and synthesis of enzyme inhibitors involved in various diseases, including cancer. In the 1990s Luis Moroder and Robert Huber supported Nediljko Budisa in establishing genetic code engineering in Germany - a research area that merges chemical syntheses with biological complexities in the form of chemical synthetic biology (Xenobiology).
Like all sugar compounds, honey caramelizes if heated sufficiently, becoming darker in color, and eventually burns. However, honey contains fructose, which caramelizes at lower temperatures than glucose. The temperature at which caramelization begins varies, depending on the composition, but is typically between 70 and 110 °C (158 and 230 °F). Honey also contains acids, which act as catalysts for caramelization. The specific types of acids and their amounts play a primary role in determining the exact temperature. Of these acids, the amino acids, which occur in very small amounts, play an important role in the darkening of honey. The amino acids form darkened compounds called melanoidins, during a Maillard reaction. The Maillard reaction occurs slowly at room temperature, taking from a few to several months to show visible darkening, but speeds up dramatically with increasing temperatures. However, the reaction can also be slowed by storing the honey at colder temperatures. Unlike many other liquids, honey has very poor thermal conductivity of 0.5 W/(m⋅K) at 13% water content (compared to 401 W/(m⋅K) of copper), taking a long time to reach thermal equilibrium. Due to its high kinematic viscosity honey does not transfer heat through momentum diffusion (convection) but rather through thermal diffusion (more like a solid), so melting crystallized honey can easily result in localized caramelization if the heat source is too hot or not evenly distributed. However, honey takes substantially longer to liquefy when just above the melting point than at elevated temperatures.

=== Modeling === In fasting volunteers, blood levels of ethanol increase proportionally with the dose of ethanol administered. Peak blood alcohol concentrations may be estimated by dividing the amount of ethanol ingested by the body weight of the individual and correcting for water dilution. For time-dependent calculations, Swedish professor Erik Widmark developed a model of alcohol pharmacokinetics in the 1920s. The model corresponds to a single-compartment model with instantaneous absorption and zero-order kinetics for elimination. The model is most accurate when used to estimate BAC a few hours after drinking a single dose of alcohol in a fasted state, and can be within 20% CV of the true value. It is less accurate for BAC levels below 0.2 g/L (alcohol is not eliminated as quickly as predicted) and consumption with food (overestimating the peak BAC and time to return to zero).
=== Ribosome stapling === The orthogonal ribosomes described above all focus on optimizing the 16S rRNA. Thus far, this optimized 16S rRNA was combined with natural large-subunits to form orthogonal ribosomes. If the 23S rRNA, the main RNA-component of the large ribosomal subunit, is to be optimized as well, it had to be assured, that there was no crosstalk in the assembly of orthogonal and natural ribosomes (see figure B). To ensure that optimized 23S rRNA would only form into ribosomes with the optimized 16S rRNA, the two rRNAs were combined into one transcript. By inserting the sequence for the 23S rRNA into a loop-region of the 16S rRNA sequence, both subunits still adopt functioning folds. Since the two rRNAs are linked and thus in constant proximity, they preferably bind each other, not other free floating ribosomal subunits.
=== Advancements in fluid flow analysis === Inviscid flow was further analyzed by various mathematicians (Jean le Rond d'Alembert, Joseph Louis Lagrange, Pierre-Simon Laplace, Siméon Denis Poisson) and viscous flow was explored by a multitude of engineers including Jean Léonard Marie Poiseuille and Gotthilf Hagen. Further mathematical justification was provided by Claude-Louis Navier and George Gabriel Stokes in the Navier–Stokes equations, and boundary layers were investigated (Ludwig Prandtl, Theodore von Kármán), while various scientists such as Osborne Reynolds, Andrey Kolmogorov, and Geoffrey Ingram Taylor advanced the understanding of fluid viscosity and turbulence.

It is a glucagon-like peptide-1 receptor agonist, often grouped with the incretin mimetics. Its backbone is modified from the human hormone to resist enzymatic degradation and to bind albumin. These two features distinguish it from the native peptide.
No. It shares high sequence identity but carries non-natural modifications that alter its stability and clearance. The naturally occurring hormone is broken down within minutes, while the synthetic analogue circulates far longer.
Attachment to serum albumin reduces renal filtration of the peptide and shields it from enzymatic breakdown. The consequence is a much longer interval between administrations than unmodified peptides allow. The precise contribution of binding affinity to overall duration is still being quantified.
The lyophilized powder is dissolved in a suitable solvent, often sterile water or a buffered diluent, with gentle mixing rather than vigorous shaking. Foaming and shear should be avoided because they can promote aggregation. The resulting solution is then stored cold and protected from light.
