Semaglutide is a synthetic peptide analog of glucagon-like peptide-1 (GLP-1), a hormone released from intestinal L-cells after food intake. The compound belongs to the incretin mimetic class and acts at GLP-1 receptors distributed across pancreatic, gastrointestinal, cardiovascular, and central nervous system tissues. Compared with native GLP-1, the molecule carries structural changes that extend its activity from minutes to roughly one week. It is studied for glycemic control in type 2 diabetes and for weight management, and its effects on cardiovascular and other outcomes remain active research areas.
Receptor binding triggers G protein signaling that raises intracellular cyclic AMP in pancreatic beta cells. Insulin release follows in a glucose-dependent manner, so secretion increases when blood glucose is elevated and diminishes when it is not. The same signaling suppresses glucagon release from alpha cells and slows gastric emptying, which blunts the post-meal glucose rise. In the brain, receptor activation in regions such as the arcuate nucleus is associated with reduced appetite and lower energy intake. How much each of these effects contributes to overall weight change is not fully settled.

The primary target is the GLP-1 receptor, a class B G protein-coupled receptor expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises intracellular cyclic AMP, which potentiates glucose-dependent insulin secretion and lowers glucagon release when blood glucose is elevated. Signalling in the hypothalamus and brainstem is associated with reduced appetite and slower gastric emptying. Because the insulinotropic effect depends on prevailing glucose levels, the hypoglycaemic risk of the peptide alone is described as low in most study settings. The relative contribution of peripheral and central actions remains an active research question.
Large randomised trials in adults with type 2 diabetes and in adults with obesity have reported reductions in body weight and improvements in several cardiovascular risk markers. One outcome trial found a lower incidence of major adverse cardiovascular events in participants with diabetes and established cardiovascular disease. Gastrointestinal effects such as nausea and vomiting are the most frequently reported adverse events and often diminish over time. Changes in lean body mass during weight loss are an area of ongoing investigation. Effects in adolescents and in pregnancy are less well characterised, and current labelling advises against use during pregnancy.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released after nutrient intake. The molecule contains 31 amino acid residues and differs from the native sequence at several positions. A non-natural residue at position eight resists the enzyme that normally truncates the hormone, while a lysine-linked fatty diacid side chain promotes binding to serum albumin. These two modifications extend the circulating half-life from minutes to roughly one week. The peptide is produced by solid-phase synthesis followed by selective acylation, and its identity and purity are confirmed by spectrometric and chromatographic techniques.
Lyophilized semaglutide powder is typically held at minus twenty degrees Celsius for long-term storage. At that temperature, solid-state degradation reactions proceed slowly and the peptide remains intact for extended periods. Repeated freeze-thaw cycles are best avoided because they promote aggregation and can shift the proportion of monomeric peptide present. Working aliquots are often prepared so that each portion is thawed only once, and desiccant is placed inside the vial to limit moisture uptake.
Identity and purity are usually assessed by reverse-phase high-performance liquid chromatography coupled to mass spectrometry. Retention time and observed mass are compared against a reference standard run under identical conditions. Impurity profiles reveal deamidation products, oxidized methionine variants, and truncated fragments that arise during synthesis or storage. Peptide mapping through enzymatic digestion confirms the primary sequence, while amino acid analysis offers an independent check on overall composition.
Stability studies examine how temperature, pH, and moisture influence degradation rates over time. In aqueous solution, hydrolysis and deamidation accelerate as pH moves away from mildly acidic conditions. Light exposure and residual metal ions can also trigger oxidation of susceptible residues. Accelerated aging at elevated temperature is used to estimate shelf life, though extrapolation to room temperature carries uncertainty because individual degradation pathways do not always scale predictably.
Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.
Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
Receptor activation follows the canonical Gs pathway: binding increases intracellular cyclic AMP, which promotes protein kinase A activity. In pancreatic beta cells this amplifies glucose-dependent insulin release, so secretion rises when blood glucose is high and changes little when it is low. The same signalling suppresses glucagon release from alpha cells and slows gastric emptying. Receptors in the hypothalamus and brainstem are thought to contribute to reduced appetite and lower energy intake. Which of these effects dominates clinical outcomes remains an area of active study.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released by intestinal L cells after food intake. The natural hormone acts on pancreatic and central receptors but is degraded within minutes by dipeptidyl peptidase-4 and other peptidases. Semaglutide belongs to the class of long-acting GLP-1 receptor agonists, a group distinguished by structural changes that slow breakdown and extend circulation time. Its development followed earlier short-acting analogues and reflects a general strategy in peptide drug design: preserve receptor activity while blocking proteolytic clearance.
Three structural changes define the molecule. At position 8 an alpha-aminoisobutyric acid residue replaces alanine, which blocks dipeptidyl peptidase-4 cleavage. At position 34 arginine replaces lysine, and at position 26 a lysine carries a C18 fatty diacid attached through a short linker. The fatty chain binds serum albumin, and this albumin association reduces renal filtration and enzymatic attack. The unchanged backbone retains the receptor contacts that produce signalling. The free base has the formula C187H291N45O59 and a molecular weight near 4114 daltons.
== Biosynthesis == The various forms of MSH are generated from different cleavages of the proopiomelanocortin protein, which also yields other important neuropeptides like adrenocorticotropic hormone. Melanocytes in skin make and secrete MSH in response to ultraviolet light, where it increases synthesis of melanin. Some neurons in arcuate nucleus of the hypothalamus make and secrete α-MSH in response to leptin; α-MSH is also made and secreted in the anterior lobe of the pituitary gland.
== Prevention == Methods in preventing amputation, limb-sparing techniques, depend on the problems that might cause amputations to be necessary. Chronic infections, often caused by diabetes or decubitus ulcers in bedridden patients, are common causes of infections that lead to gangrene, which, when widespread, necessitates amputation. There are two key challenges: first, many patients have impaired circulation in their extremities, and second, they have difficulty curing infections in limbs with poor blood circulation. Crush injuries where there is extensive tissue damage and poor circulation also benefit from hyperbaric oxygen therapy (HBOT). The high level of oxygenation and revascularization speed up recovery times and prevent infections. A study found that the patented method called Circulator Boot achieved significant results in prevention of amputation in patients with diabetes and arteriosclerosis. Another study found it also effective for healing limb ulcers caused by peripheral vascular disease. The boot checks the heart rhythm and compresses the limb between heartbeats; the compression helps cure the wounds in the walls of veins and arteries, and helps to push the blood back to the heart. For victims of trauma, advances in microsurgery in the 1970s have made replantation of severed body parts possible. The establishment of laws, rules, and guidelines, and the employment of modern equipment help protect people from traumatic amputations.
Sachets, canisters, pads, cards, and emitters Labels and printed indicators Bottle caps and closures Polymer films and multilayer laminates Paper, board, plastic, glass, and metal coatings Trays and moulded package components Adhesives and absorbent structures Electronic tags, sensors, and data loggers The active component may be positioned inside a package without directly touching the product. It may also be immobilised on a food-contact surface or incorporated into the packaging material. System design must consider the quantity of active material, release or absorption rate, package volume, barrier performance, product composition, storage temperature, expected shelf life, migration limits, consumer safety, manufacturing conditions, recycling, and disposal. Edible films, bio-based polymers, biodegradable materials, nanoparticles, natural antimicrobials, and responsive colour indicators are continuing areas of research. Silver nanoparticles and other antimicrobial nanomaterials have been studied for packaging applications, but their migration, toxicity, regulatory status, recycling impact, and environmental effects require evaluation.

== Structure == The 3D structure of human serum albumin has been determined by X-ray crystallography to a resolution of 2.5 ångströms (250 pm). Albumin is a 65–70kDa protein. Albumin comprises three homologous domains that assemble to form a heart-shaped protein. Each domain is a product of two subdomains that possess common structural motifs. The principal regions of ligand binding to human serum albumin are located in hydrophobic cavities in subdomains IIA and IIIA, which exhibit similar chemistry. Structurally, the serum albumins are similar, each domain containing five or six internal disulfide bonds.
Zverev's first ATP event of the year was the 2016 Australian Open, where he had a difficult draw against world No. 2 Andy Murray and was only able to win six games in his opening match. He bounced back at the Open Sud de France, reaching the semifinals in the singles event and his second career final with his brother in the doubles event. During the singles event, he upset No. 13 Marin Čilić, the highest-ranked player he had defeated in an official match at the time. Zverev then produced a strong result at the Indian Wells Masters, where he defeated two top 30 players. In the fourth round, he nearly upset No. 5 Rafael Nadal. Zverev had a match point while he was serving for the final set, but missed a routine forehand volley and then proceeded to lose 14 of the remaining 15 points in the match. He commented on the match point that, "I missed probably the easiest shot I had the whole match." Back in Europe, Zverev was able to recover as he reached his first ATP singles final at the Open de Nice Côte d'Azur, finishing runner-up to No. 15 Dominic Thiem, the top seed and defending champion. Thiem also defeated him in the third round of the French Open.
Cossack relations with the Tsardom of Russia were varied from the outset. At times they supported Russian military operations, at other times they rebelled against the central power. After one such uprising at the end of the 18th century, Russian forces destroyed the Zaporozhian Host. Many of the Cossacks who had remained loyal to the Russian Monarch and continued their service later moved to the Kuban. Others, choosing to continue a mercenary role, escaped control in the large Danube Delta. The service of the Cossacks in the Napoleonic wars led them to be celebrated as Russian folk heroes, and throughout the 19th century a "powerful myth" was promoted by the government that portrayed the Cossacks as having a special and unique bond to the Emperor. This image of the Cossacks as ultra-patriotic defenders of not only Russia, but also of the House of Romanov was embraced by many ordinary Cossacks, making them into a force for conservatism. By the 19th century, the Russian Empire had annexed the territory of the Cossack Hosts, and controlled them by providing privileges for their service such as exemption from taxation and allowing them to own the land they farmed. At this time, the Cossacks served as military forces in many wars conducted by the Russian Empire. Cossacks were considered excellent for scouting and reconnaissance duties, and for ambushes. Their tactics in open battle were generally inferior to those of regular soldiers, such as the Dragoons.
=== Repair of Oxidative damage === Secondary defenses include DNA-repair systems, proteolytic and lipolytic enzymes. DNA repair enzymes include endonuclease IV, induced by oxidative stress, and exonuclease III, induced in the stationary phase and in starving cells. These enzymes act on duplex DNA and clean up DNA 3' terminal ends. Prokaryotic cells contain catalysts that modify the primary structure of proteins frequently by reducing disulfide bonds. This occurs in the following steps: (i) thioredoxin reductase transfers electrons from NADPH to thioredoxin via a flavin carrier (ii) glutaredoxin is also able to reduce disulfide bonds, but using GSH as an electron donor (iii) protein disulfide isomerase facilitates disulfide exchange reactions with large inactive protein substrates, besides having chaperone activity Oxidation of surface exposed methionine residues surrounding the entrance to the active site could function as a “last-chance” antioxidant defense system for proteins.

=== Sensory and nervous systems === Although starfish do not have many well-defined sense organs, they do perceive touch, light, temperature, orientation and the status of the water around them. The tube feet, spines and pedicellariae are sensitive to touch. The tube feet, especially those at the tips of the rays, are also sensitive to chemicals, enabling the starfish to detect odour sources such as food. There are eyespots at the ends of the arms, each one made of 80–200 simple ocelli composed of pigmented epithelial cells. Individual photoreceptor cells are present in other parts of their bodies and respond to light. Whether they advance or retreat depends on the species. Traditionally, starfish were thought to lack a centralized brain. Instead, there is a complex nervous system with a nerve ring around the mouth and a radial nerve running along the ambulacral region of each arm parallel to the radial canal. The peripheral nerve system consists of two nerve nets: one in the epidermis and the other in the lining of the coelomic cavity, which are the sensory and motor systems respectively. Neurons passing through the dermis join the two. Both the ring and radial nerves function in movement and sensory. The sensory component is supplied with information from the sensory organs while the motor nerves control the tube feet and musculature. If one arm detects something attractive, it becomes dominant and temporarily over-rides the other arms to initiate movement towards it. Recent genetic evidence, however, suggests that the majority of an echinoderm's body is "all-brain".
It is afternoon in a French garden. Three men are sitting at a table, two of them playing cards while the third smokes and reads a newspaper. The man who is not playing cards calls over a young girl and has her fetch a woman with a bottle of beer. He proceeds to pour glasses for himself and his friends. After drinking the beer, the man reads a story out of the newspaper, and his friends laugh.
== Mechanism of action == The burning and painful sensations associated with capsaicin result from "defunctionalization" of nociceptor nerve fibers by causing a topical hypersensitivity reaction in the skin. As a member of the vanilloid family, capsaicin binds to a receptor on nociceptor fibers called the vanilloid receptor subtype 1 (TRPV1). TRPV1, which can also be stimulated with heat, protons, and physical abrasion, permits cations to pass through the cell membrane when activated. The resulting depolarization of the neuron stimulates it to send impulses to the brain. By binding to TRPV1 receptors, capsaicin produces similar sensations to those of excessive heat or abrasive damage, such as warming, tingling, itching, or stinging, explaining why capsaicin is described as an irritant on the skin and eyes or by ingestion. Clarifying the mechanisms of capsaicin effects on skin nociceptors was part of awarding the 2021 Nobel Prize in Physiology or Medicine, as it led to the discovery of skin sensors for temperature and touch, and identification of the single gene causing sensitivity to capsaicin.

Native GLP-1 is degraded within minutes by dipeptidyl peptidase-4 and cleared quickly. Semaglutide carries a position 8 substitution that blocks that cleavage and a fatty diacid chain that binds albumin. Together these changes extend its circulating half-life to about one week.
