The semaglutide sequence is a 31-residue analogue of human GLP-1, altered at three positions relative to the parent hormone. Aminoisobutyric acid replaces alanine at position 8, arginine replaces lysine at position 34, and a lipophilic diacid is attached to lysine 26 through a short linker. These features are reported consistently in the structural literature. The position 8 substitution blocks recognition by dipeptidyl peptidase-4, while the attached chain drives strong, reversible association with a carrier protein in blood.
Receptor activation raises intracellular cyclic AMP through Gs coupling, which promotes glucose-dependent insulin release and suppresses glucagon secretion when blood glucose is elevated. Effects outside the pancreas include slower gastric emptying and altered appetite signalling in the hypothalamus and hindbrain. The relative contribution of each tissue to overall metabolic outcomes remains an area of active investigation. Central mechanisms in particular are inferred mainly from animal models and indirect human measures rather than direct observation.
Serum protein binding dominates the pharmacokinetic profile. The attached chain associates strongly with albumin, shielding the peptide from enzymatic attack and slowing filtration by the kidney. This interaction extends the circulation half-life to roughly one week in humans, which supports weekly administration intervals. An oral version pairs the peptide with an absorption enhancer that transiently alters gastric epithelium, permitting limited uptake; bioavailability by that route is substantially lower than by injection.

Three structural changes define the molecule. At position 8 an alpha-aminoisobutyric acid residue replaces alanine, which blocks dipeptidyl peptidase-4 cleavage. At position 34 arginine replaces lysine, and at position 26 a lysine carries a C18 fatty diacid attached through a short linker. The fatty chain binds serum albumin, and this albumin association reduces renal filtration and enzymatic attack. The unchanged backbone retains the receptor contacts that produce signalling. The free base has the formula C187H291N45O59 and a molecular weight near 4114 daltons.
Receptor activation follows the canonical Gs pathway: binding increases intracellular cyclic AMP, which promotes protein kinase A activity. In pancreatic beta cells this amplifies glucose-dependent insulin release, so secretion rises when blood glucose is high and changes little when it is low. The same signalling suppresses glucagon release from alpha cells and slows gastric emptying. Receptors in the hypothalamus and brainstem are thought to contribute to reduced appetite and lower energy intake. Which of these effects dominates clinical outcomes remains an area of active study.
Large randomised trials in adults with type 2 diabetes and in adults with obesity have reported reductions in body weight and improvements in several cardiovascular risk markers. One outcome trial found a lower incidence of major adverse cardiovascular events in participants with diabetes and established cardiovascular disease. Gastrointestinal effects such as nausea and vomiting are the most frequently reported adverse events and often diminish over time. Changes in lean body mass during weight loss are an area of ongoing investigation. Effects in adolescents and in pregnancy are less well characterised, and current labelling advises against use during pregnancy.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released after nutrient intake. The molecule contains 31 amino acid residues and differs from the native sequence at several positions. A non-natural residue at position eight resists the enzyme that normally truncates the hormone, while a lysine-linked fatty diacid side chain promotes binding to serum albumin. These two modifications extend the circulating half-life from minutes to roughly one week. The peptide is produced by solid-phase synthesis followed by selective acylation, and its identity and purity are confirmed by spectrometric and chromatographic techniques.
The primary target is the GLP-1 receptor, a class B G protein-coupled receptor expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises intracellular cyclic AMP, which potentiates glucose-dependent insulin secretion and lowers glucagon release when blood glucose is elevated. Signalling in the hypothalamus and brainstem is associated with reduced appetite and slower gastric emptying. Because the insulinotropic effect depends on prevailing glucose levels, the hypoglycaemic risk of the peptide alone is described as low in most study settings. The relative contribution of peripheral and central actions remains an active research question.
=== New ownership === In December 2017, Patrick Cryne and his family sold an 80% stake in the club to NewCity Capital's Chien Lee and Pacific Media Group's Paul Conway. They were joined by Indian investor Neerav Parekh and Billy Beane, minority owner of and advisor for the Athletics, who was widely known for the Moneyball film portrayal, as part of an international investor consortium. Barnsley were relegated to the third tier in 2017–18. Following this, the new owners adopted a data-driven approach to identify talents, focusing on young players and team rebuilding. The club appointed Daniel Stendel as head coach, who introduced a high-pressing style of play. Barnsley were promoted back to the Championship the following season. In the 2019–20 season, under new coach Gerhard Struber, Barnsley avoided relegation from the Championship. In 2020–21, under the management of Valérien Ismaël, Barnsley finished fifth place and reached the EFL Championship Play-offs for the first time in 24 years, doing so with the youngest squad and one of the smallest budgets in the division. The Wall Street Journal called Barnsley a "Moneyball experiment". Prior to the 2021–22 season, Markus Schopp was appointed as the new head coach. In November 2021, he was dismissed after seven consecutive defeats. Three weeks later Poya Asbaghi was appointed as his successor. Fortunes improved little as Barnsley were relegated from the 2021–22 EFL Championship following a 2–1 defeat against Huddersfield Town. Asbaghi left the club by mutual consent shortly afterwards.
However, these authors also found that MOR244-3 lacks the specific metal ion binding site suggested by Suslick, instead showing a different motif in the EC2 domain. Malfunction of the metalloproteins in the olfactory system is hypothesized to have a connection with amyloidal based neurodegenerative diseases.
== Adorno translated into English == While even German readers can find Adorno's work difficult to understand, an additional problem for English readers is that his German idiom is particularly difficult to translate into English. A similar difficulty of translation is true of Hegel, Heidegger, and a number of other German philosophers and poets. As a result, some early translators tended toward over-literalness. Recently, Edmund Jephcott and Stanford University Press have published new translations of some of Adorno's lectures and books, including Introduction to Sociology, Problems of Moral Philosophy, his transcribed lectures on Kant's Critique of Pure Reason and Aristotle's "Metaphysics," and a new translation of the Dialectic of Enlightenment. Professor Henry Pickford, of the University of Colorado at Boulder, has translated many of Adorno's works, such as "The Meaning of Working Through the Past." A new translation has also appeared of Aesthetic Theory and the Philosophy of New Music by Robert Hullot-Kentor from the University of Minnesota Press. Hullot-Kentor is also currently working on a new translation of Negative Dialectics. Adorno's correspondence with Alban Berg, Towards a Theory of Musical Reproduction, and the letters to Adorno's parents have been translated by Wieland Hoban and published by Polity Press. These fresh translations are slightly less literal in their rendering of German sentences and words and are more accessible to English readers.

Coenzyme Q (CoQ ), also known as ubiquinone, is a naturally occurring biochemical cofactor (coenzyme) and an antioxidant produced by the human body. The human body mainly produces the form known as coenzyme Q10 (CoQ10, ubidecarenone), but other forms exist. CoQ is used by and found in many organisms, including animals and bacteria. As a result, it can also be obtained from dietary sources, such as meat, fish, seed oils, vegetables, and dietary supplements. CoQ plays a role in mitochondrial oxidative phosphorylation, aiding in the production of adenosine triphosphate (ATP), which is involved in energy transfer within cells. The structure of CoQ10 consists of a benzoquinone moiety and an isoprenoid side chain, with the "10" referring to the number of isoprenyl chemical subunits in its tail. Although a ubiquitous molecule in human tissues, CoQ10 is not a dietary nutrient and does not have a recommended intake level, and its use as a supplement is not approved in the United States for any health or anti-disease effect.
The most important transport way in Schleswig-Holstein is Kiel Canal, which connect Brunsbüttel on North Sea with Kiel on Baltic Sea. Total cargo of ships reach peaks in 2007 and 2012, after that it continuous decline with 73.8 million tonnes in 2020.
The precursor to indigo is indican, a colorless, water-soluble derivative of the amino acid tryptophan, and Indigofera leaves contain as much as 0.2–0.8% of this compound. Pressing cut leaves into a vat and soaking hydrolyzes the indican, releasing β-D-glucose and indoxyl. The indoxyl dimerizes in the mixture, and after 12–15 hours of fermentation yields the yellow, water-soluble leucoindigo. Subsequent exposure to air forms the blue, water-insoluble indigo dye. The dye precipitates from the fermented leaf solution upon oxidation, but may also be precipitated when mixed with a strong base such as lye. The solids are filtered, pressed into cakes, dried, and powdered. The powder is then mixed with various other substances to produce different shades of blue and purple. Natural sources of indigo also include mollusks: the Murex genus of sea snails produces a mixture of indigo and 6,6'-dibromoindigo (red), which together produce a range of purple hues known as Tyrian purple. Light exposure during part of the dyeing process can convert the dibromoindigo into indigo, resulting in blue hues known as royal blue, hyacinth purple, or tekhelet.
Because trans fats are more linear, they crystallize more easily, allowing them to be solid (rather than liquid) at room temperatures. This has several processing and storage advantages. In nature, unsaturated fatty acids generally have cis configurations as opposed to trans configurations. Saturated fatty acids (those without any carbon-carbon double bonds) are abundant (see tallow), but they also can be generated from unsaturated fats by the process of fat hydrogenation. In the course of hydrogenation, some cis double bonds convert into trans double bonds. Chemists call this conversion an isomerization reaction. Any molecule with a C=C double bond can be either a trans or a cis fatty acid depending on the configuration of the double bond. For example, oleic acid and elaidic acid are both unsaturated fatty acids with the chemical formula C9H17C9H17O2. They both have a double bond located midway along the carbon chain. It is the geometry of this bond that sets oleic and elaidic acids apart. They have distinct physical-chemical properties of the molecule. For example, the melting point of elaidic acid is 45 °C, which is higher than that of oleic acid. This notably means that it is a solid at human body temperatures.

==== Tolerance ==== Tolerance is a process characterized by neuroadaptations that result in reduced drug effects. While receptor upregulation may often play an important role other mechanisms are also known. Tolerance is more pronounced for some effects than for others; tolerance occurs slowly to the effects on mood, itching, urinary retention, and respiratory depression, but occurs more quickly to the analgesia and other physical side effects. However, tolerance does not develop to constipation or miosis (the constriction of the pupil of the eye to less than or equal to two millimeters). This idea has been challenged, however, with some authors arguing that tolerance does develop to miosis. Tolerance to opioids is attenuated by a number of substances, including:
== History == Following the discovery in 1952 that the tuberculosis drug iproniazid elevated the mood of people taking it, and the subsequent discovery that the effect was likely due to inhibition of monoamine oxidase (MAO) and elevation of monoamine neurotransmitters in the brain, many people and companies started trying to discover monoamine oxidase inhibitors (MAOIs) to use as antidepressants. Deprenyl, the racemic form of selegiline, was synthesized and discovered by Zoltan Ecseri at the Chinoin Pharmaceutical Company (part of Sanofi since 1993) in Budapest, Hungary. Chinoin received a patent on the drug in 1962 and the compound was first published in the scientific literature in English in 1965. Chinoin researchers had been studying substituted amphetamines since 1960, and decided to try synthesizing amphetamines that acted as MAOIs. It had been known that methamphetamine was a reversible inhibitor of MAO. Deprenyl, also known as N-propargyl-N-methylamphetamine, is closely related to and inspired by pargyline (N-propargyl-N-methylbenzylamine), another MAOI that had been synthesized earlier. Deprenyl was initially referred to by the chemical name phenylisopropylmethylpropinylamine and the developmental code name E-250. Work on the biology and effects of E-250 in animals and humans was conducted by a group led by József Knoll at Semmelweis University, which was also in Budapest. Deprenyl is a racemic compound (a mixture of two isomers called enantiomers).
==== Limitations ==== Due to the necessity of oxygen for PDT, these treatments do not work as well in hypoxic environments, including in developed tumors and some deep wounds. Dental infections tend to also respond better to photothermal therapy than photodynamic therapy, though both have a strong effect. The efficacy of PDT for antimicrobial usage is limited by the properties of the membrane of the target cell such as the electrical gradient (membrane potential) and lipid composition. Whereas high cell death is observed for Escherichia coli and Staphylococcus aureus, other bacterial species such as Klebsiella pneumoniae and Acinetobacter baumannii tend to see very low impact from PDT due to these factors. This limits potential as a broadband antibiotic, but may also allow for specificity in targeting the pathogenic cells over human and skin microbiome cells.
=== Extra-articular === JIA is associated with inflammation in the front of the eye (acute or chronic anterior uveitis), which affects about one in six children with JIA. Eye involvement occurs most commonly in girls, those with only a few joints involved (oligoarthritis), those with rheumatoid factor negative polyarticular JIA, and those with a positive anti-nuclear antibody (ANA). Girls younger than six years old are at greatest risk of anterior uveitis. 90% of chronic anterior uveitis occurs within 4 years of JIA diagnosis. Acute anterior uveitis is seen in 11-13% of JIA and is more commonly associated with the enthesitis related arthritis subtype. It presents with light sensitivity, redness of the eyes and eye pain. Regular eye exams are required for children affected by acute or chronic anterior uveitis related to JIA. Children with the Systemic JIA subtype often experience extra-articular manifestations including fever, rash, enlarged lymph nodes, enlarged liver or spleen, serositis and anemia.

It is a synthetic peptide of 31 amino acids, built to resemble the natural incretin hormone GLP-1. Because of its size and composition it is handled analytically like other therapeutic peptides, using chromatographic and mass spectrometric methods rather than the techniques typical of small organic drugs.
