Three structural changes define the molecule. At position 8 an alpha-aminoisobutyric acid residue replaces alanine, which blocks dipeptidyl peptidase-4 cleavage. At position 34 arginine replaces lysine, and at position 26 a lysine carries a C18 fatty diacid attached through a short linker. The fatty chain binds serum albumin, and this albumin association reduces renal filtration and enzymatic attack. The unchanged backbone retains the receptor contacts that produce signalling. The free base has the formula C187H291N45O59 and a molecular weight near 4114 daltons.
Receptor activation follows the canonical Gs pathway: binding increases intracellular cyclic AMP, which promotes protein kinase A activity. In pancreatic beta cells this amplifies glucose-dependent insulin release, so secretion rises when blood glucose is high and changes little when it is low. The same signalling suppresses glucagon release from alpha cells and slows gastric emptying. Receptors in the hypothalamus and brainstem are thought to contribute to reduced appetite and lower energy intake. Which of these effects dominates clinical outcomes remains an area of active study.
Lyophilized semaglutide is typically stored at temperatures between minus 20 and minus 80 degrees Celsius for long-term preservation. Short-term storage at 2 to 8 degrees Celsius is common for working aliquots. Repeated freeze-thaw cycles can degrade the peptide and are usually avoided. The molecule is hygroscopic in its solid form, so containers should remain sealed with desiccant. Solutions are less stable than powders and are generally prepared fresh. Light exposure is limited because aromatic residues can undergo photo-oxidation.
Semaglutide dissolves readily in water and in aqueous buffers near neutral pH. Solubility decreases near the isoelectric point, where net charge is minimal. Common laboratory solvents include phosphate-buffered saline and dilute ammonium bicarbonate. Strongly acidic or basic conditions may accelerate hydrolysis. Working concentrations are usually prepared by diluting a concentrated stock. Vial surfaces can adsorb small amounts of peptide at low concentrations, so carrier proteins or low-binding tubes are sometimes used.
Reverse-phase high-performance liquid chromatography is the standard method for purity assessment, separating the peptide from truncated or oxidized variants. Mass spectrometry confirms molecular mass and detects modifications, while ultraviolet absorbance near 280 nanometers supports concentration measurement through tryptophan and tyrosine residues. Circular dichroism can indicate secondary structure, though the peptide is largely helical in solution, and ion-exchange chromatography resolves charge variants. Purity values above 95 percent are typical for research-grade material. Stability studies track degradation over time under defined conditions.

Pharmacological activity arises from agonism at the glucagon-like peptide-1 receptor, a G protein-coupled receptor expressed in the pancreas, the gastrointestinal tract, and the brainstem. Receptor activation raises intracellular cyclic adenosine monophosphate and enhances insulin release in a glucose-dependent manner, an effect that diminishes when blood glucose concentration is low. Other effects include slowed gastric emptying and hypothalamic satiety signalling. These pathways are described well. Receptor desensitisation rates across tissues, relative to the endogenous hormone, are still under investigation, and reported findings differ between laboratories.
The company that developed the compound filed it as a long-acting analogue, and it gained first approval in 2017 for type 2 diabetes. Later authorisations from several regulators extended the indication to chronic weight management, and the World Health Organization added the glucagon-like peptide-1 receptor agonist drug class to its model list of essential medicines in 2023. Production uses solid-phase peptide synthesis followed by side-chain conjugation and chromatographic purification. Supply constraints and cost differences across regions are well documented. Literature on long-term outcomes continues to grow, with many trials reporting surrogate endpoints rather than hard clinical endpoints.
Semaglutide is a synthetic peptide of thirty-one amino acids that shares roughly ninety-four percent sequence identity with human glucagon-like peptide-1. Two substitutions resist enzymatic cleavage by dipeptidyl peptidase-4, and a fatty diacid side chain attached through a linker promotes binding to serum albumin. That albumin binding slows renal clearance and extends the circulating half-life from minutes to approximately one week. The structural changes are well established in the published literature. Whether the same modifications affect receptor signalling bias in ways that matter clinically remains an open question.
== Sustainable development goals == According to Project Breakthrough, digital agriculture can help advance the United Nations Sustainable Development Goals by providing farmers with more real-time information about their farms, allowing them to make better decisions. Technology improves crop production by providing a better understanding of soil health. It allows farmers to use fewer pesticides on their crops. Soil and weather monitoring reduces water waste. Digital agriculture can lead to economic growth by allowing farmers to get the most production out of their land. The loss of agricultural jobs can be offset by new job opportunities in manufacturing and maintaining the necessary technology for the work. Digital agriculture also enables individual farmers to work in concert, collecting and sharing data using technology.
The 20S proteasome is both ubiquitous and essential in eukaryotes and archaea. The bacterial order Actinomycetales, also share homologs of the 20S proteasome, whereas most bacteria possess heat shock genes hslV and hslU, whose gene products are a multimeric protease arranged in a two-layered ring and an ATPase. The hslV protein has been hypothesized to resemble the likely ancestor of the 20S proteasome. In general, HslV is not essential in bacteria, and not all bacteria possess it, whereas some protists possess both the 20S and the hslV systems. Many bacteria also possess other homologs of the proteasome and an associated ATPase, most notably ClpP and ClpX. This redundancy explains why the HslUV system is not essential. Sequence analysis suggests that the catalytic β subunits diverged earlier in evolution than the predominantly structural α subunits. In bacteria that express a 20S proteasome, the β subunits have high sequence identity to archaeal and eukaryotic β subunits, whereas the α sequence identity is much lower. The presence of 20S proteasomes in bacteria may result from lateral gene transfer, while the diversification of subunits among eukaryotes is ascribed to multiple gene duplication events.

=== Patent history === The U.S. Food and Drug Administration (FDA) approved tramadol in March 1995, and an extended-release (ER) formulation in September 2005. ER Tramadol was protected by US patents nos. 6,254,887 and 7,074,430. The FDA listed the patents' expiration as 10 May 2014. However, in August 2009, the US District Court for the District of Delaware ruled the patents invalid, a decision upheld the following year by the Court of Appeals for the Federal Circuit. Manufacture and distribution of generic equivalents of Ultram ER in the United States was therefore permitted before the expiration of the patents.

Hamish de Bretton-Gordon, a British chemical weapons expert and former commanding officer of the UK's Joint Chemical, Biological, Radiation and Nuclear Regiment and its NATO equivalent, "dismissed" suggestions that Novichok agents could be found in other places in the former Soviet Union such as Uzbekistan and has asserted that Novichok agents were produced only at Shikhany in Saratov Oblast, Russia. Mirzayanov also says that it was at Shikhany, in 1973, that scientist Pyotr Petrovich Kirpichev first produced Novichok agents; Vladimir Uglev joined him on the project in 1975. According to Mirzayanov, while production took place in Shikhany, the weapon was tested at Nukus between 1986 and 1989. Following the poisoning of the Skripals, former head of the GosNIIOKhT security department Nikolay Volodin confirmed in an interview to Novaya Gazeta that there have been tests at Nukus, and said that dogs were used. In May 2018, the Irish Independent reported that "Germany's foreign intelligence service secured a sample of the Soviet-developed nerve agent Novichok in the 1990s and passed on its knowledge to partners including Britain and the US, according to German media reports." The sample was analysed in Sweden. Small amounts of the Novichok nerve agent were subsequently produced in some NATO countries for test purposes.

According to Clayton Thyne and Jonathan Powell's coup data set, there were 457 coup attempts from 1950 to 2010, of which 227 (49.7%) were successful and 230 (50.3%) were unsuccessful. They find that coups have "been most common in Africa and the Americas (36.5% and 31.9%, respectively). Asia and the Middle East have experienced 13.1% and 15.8% of total global coups, respectively. Europe has experienced by far the fewest coup attempts: 2.6%." Most coup attempts occurred in the mid-1960s, but there were also large numbers of coup attempts in the mid-1970s and the early 1990s. From 1950 to 2010, a majority of coups failed in the Middle East and Latin America. They had a somewhat higher chance of success in Africa and Asia. Numbers of successful coups have decreased over time. A number of political science datasets document coup attempts around the world and over time, generally starting in the post-World War II period. Major examples include the Global Instances of Coups dataset, the Coups & Political Instability dataset by the Center of Systemic Peace, the Coup d'état Project by the Cline Center, the Colpus coup dataset, and the Coups and Agency Mechanism dataset. A 2023 study argued that major coup datasets tend to over-rely on international news sources to gather their information, potentially biasing the types of events included. Its findings show that while such a strategy is sufficient for gathering information on successful and failed coups, attempts to gather data on coup plots and rumors require a greater consultation of regional and local-specific sources.
The Genetics & IVF Institute (GIVF) is an international provider of infertility and genetics services and products, and also engages in biomedical research in these fields. The Institute was founded in 1984 by Dr. Joseph D. Schulman and associates. GIVF headquarters are in Fairfax, VA, US, and its facilities include locations in Pennsylvania, Minnesota, California, and Texas in the United States, as well as in China, Mexico, and several other countries. Genetics & IVF Institute has been responsible for numerous key innovations to help couples have healthy babies and to improve the reproductive health of women. These include: First U.S. medical center to introduce non-surgical ultrasound guided egg retrieval (transvaginal oocyte retrieval) for IVF; achieved America's first frozen embryo twin birth; first American team to publicly report pregnancies using ICSI (intracytoplasmic sperm injection); first to offer clinical laboratory testing for certain mutations in the BRCA1 and BRCA2 genes which cause hereditary breast cancer; developed the important technique of non-surgical sperm aspiration for improved treatment of male infertility; first in the world to initiate ovarian cryopreservation for storage of oocytes for women with cancer; discovered a method (non-disclosing preimplantation genetic testing) for the effective prevention of Huntington disease; developed MicroSort, an important flow-cytometric human sperm separation technique for preconceptual gender selection.
In some reactions, protons and hydroxide may directly act as acid and base in term of specific acid and specific base catalysis. But more often groups in substrate and active site act as Brønsted–Lowry acid and base. This is called general acid and general base theory. The easiest way to distinguish between them is to check whether the reaction rate is determined by the concentrations of the general acid and base. If the answer is yes then the reaction is the general type. Since most enzymes have an optimum pH of 6 to 7, the amino acids in the side chain usually have a pKa of 4~10. Candidate include aspartate, glutamate, histidine, cysteine. These acids and bases can stabilise the nucleophile or electrophile formed during the catalysis by providing positive and negative charges.
Contributions to island of inversion measurements and potential discovery of new magic numbers The island of inversion is a region of the chart of nuclides in which isotopes have enhanced stability, compared to the surrounding unstable nuclei. The island is associated with the magic neutron numbers (N = 8, 14, 20, 28, 50, 82, 126), where this breakdown occurs. Various experiments at ISOLDE have determined properties of these island of inversion isotopes, including the first of their kind measurements performed with Miniball on magnesium-32, lying in the island of inversion at N = 20. Furthermore, the ISOLTRAP experiment provided results using calcium-52 to reveal a potential new magic number, 32, which was later disproven by the CRIS experiment. Production of isomeric beams A nuclear isomer is a metastable state of a nucleus, in which one or more nucleons occupy higher energy levels than in the ground state of the same nucleus. In the mid-2000s, REX-ISOLDE developed a technique to select and post-accelerate isomeric beams to use in nuclear-decay experiments, such as at Miniball.
RNA sequencing (RNA-seq) RNA-seq is a high-throughput RNA sequencing technology that allows scientists to profile the entire RNA (transcriptome). Therefore, novel transcripts and gene expression level can be identified based on cDNA libraries. This method can be used for cancer diagnosis and treatment evaluation. Reverse transcription polymerase chain reaction (RT-PCR) RT-PCR is a widely used mRNA expression detection method. It enables reverse transcription of mRNA to cDNA for further identification and qualification. In early 1992, RT-PCR was applied in PSA gene expression in peripheral blood for early prostate cancer diagnosis. Digital PCR (dPCR) dPCR is a relatively accurate quantification method of measuring the initial concentration of mRNA targets. It can be applied to analyze genetic and epigenetic changes. In-situ hybridization (ISH) ISH is a tissue dependent visualization method of identifying mRNA targeted in the samples. The "tissue" can be blood sample. In chronic myeloid leukemia, ISH has been applied on peripheral-blood specimens.

Native GLP-1 is a short-lived peptide cleared within one to two minutes by dipeptidyl peptidase-4 and related enzymes. Semaglutide keeps the receptor-binding backbone but adds substitutions and a lipid chain. These changes block the main cleavage site and allow reversible albumin binding, extending the half-life to roughly 165 hours.
Albumin is the most abundant protein in plasma and carries molecules that bear fatty-acid chains. Binding shields the peptide from renal filtration and from peptidases, keeping a circulating reservoir. Slow release from this reservoir produces sustained receptor occupancy and supports infrequent dosing.
The insulinotropic effect is glucose-dependent, meaning secretion increases mainly when glucose is elevated. This property is often described as lowering the chance of hypoglycaemia when the compound is used alone. Other glucose-lowering agents used at the same time can still cause low blood glucose.
Long-term storage is usually at minus 20 to minus 80 degrees Celsius in a sealed, desiccated container. Working aliquots can be held briefly at 2 to 8 degrees Celsius.
